[Abstract] Objective: The purpose of this study is to verify the depigmented role of regimen containing niacinamide and N-acetylglucosamine (N/NAG) on facial hyperpigmentation. Methods: This study was a vehicle-controlled, full-face, parallel group comparison in Chinese female subjects with facial hyperpigmentation. All pictures were taken by REAL 3.0 system. Spot area fraction, L*a*b, NC2 melanin spot area fraction, NC2 melanin gray scale and NC2 melanin evenness were analyzed by this system. A Chromameter was used to measure the L*a*b value for non-lesion area. Expert assessment on color pictures before and after the usage and self assessment questionnaires were also used. Results: Against the vehicle control, the N/NAG regimen provided significant benefits on reducing spot area fraction, minimized skin darkening and yellowing, increased skin tone evenness and minimized the NC2 melanin gray scale. Conclusions: The N/NAG regimen was well tolerated. Against the vehicle control, it produced a significant improvement in skin color and hyperpigmentation.
